That you can do by adjusting the NTFS permissions on the
shared area and by verifying the share level permissions allow
the accounts the desired access .
If the group is to have read on the one folder, then make sure
it has read at the share level , grant the group read on the one
folder , and then make sure no group that has as members any
account in the grantee group (like Users) is used on other
folders in the shared area. The users will have access to
any part to which they have a grant no matter by what group
it has been made.

That you cannot do. In order to log in accounts need a
certain amount of access to the system files area.
