Maybe some help here...
http://download.microsoft.com/download/5/6/1/561c9fd7-0e27-4525-94ec-4d2d38f61aa3/TSHT_SBS.htm#backup
--------
Backup fails, reporting "'Script.bks' file not found."
Cause: The Exchange Information Store is not running.
Solution: Start the Exchange Information Store.
To start the Exchange Information Store
Click Start, click Run, and then type Services.msc.
In the details pane, right-click Microsoft Exchange Information Store, and
then click Start.
Cause:
A folder explicitly marked to be included in the backup is not on the
system.
The Small Business Backup Script has been deleted or is corrupted.
The UNC path you are backing up to does not exist or is inaccessible.
Solution: Re-run the Backup Configuration Wizard from the Backup snap-in in
Server Management, accepting the defaults to reset.

I have an issue with a backup failing and yes I have looked through
the
"Troubleshooting Windows Small Business Server 2003 Guide" to make sure
the
information store was running and that all the exclusion folders exist. Is
there a log to find more detailed information?
